چکیده

Early and precise pathogen identification corresponding disease management are primary concerns in aquaculture. Here, we attempted at diagnostic methods that can simultaneously identify multiple pathogens, where many samples, several concurrent infections to be handled. Hence, a multiplex PCR assay targeting five major aquaculture viz. Vibrio parahaemolyticus, anguillarum, alginolyticus, vulnificus harveyi was developed for the first time. The primers toxR of V. amiB col vvhA vulnificus, topA were applied. Furthermore, reaction included an internal amplification control against prokaryotic 16S rRNA perceive false-negative results. showed 100% specificity 56 unique bacteria. sensitivity 0.25 ng harveyi, 0.5 vulnificus, 1 parahaemolyticus 2 alginolyticus DNA per µL assay. Sensitivity regarding CFU 1.2, 5.2, 10, 5.6×101 3.8×102 µL, respectively. results suggest optimized method applied sensitive specific pathogens through single

Development and evaluation of a multiplex PCR for simultaneous detection of five foodborne pathogens.

AIMS To develop a rapid multiplex PCR method for simultaneous detection of five major foodborne pathogens (Staphylococcus aureus, Listeria monocytogenes, Escherichia coli O157:H7, Salmonella Enteritidis and Shigella flexneri, respectively). METHODS AND RESULTS Amplification by PCR was optimized to obtain high efficiency. Sensitivity and specificity assays were investigated by testing differen...

A novel magnetic capture–multiplex PCR assay for the simultaneous detection of three foodborne pathogens

doi: 10.1111/j.1757-837X.2011.00113.x Abstract Introduction Salmonella, Shigella and Escherichia coli O157: H7 are major foodborne pathogens that cause gastrointestinal diseases worldwide. Apart from food contamination, fecal pollution has been consistently associated with the transmission of these pathogens, and their rapid detection in food and stools is of significance for food safety. Howev...

Development and evaluation of a novel multiplex PCR technology for molecular differential detection of bacterial respiratory disease pathogens.

The ResPlex I assay (Qiagen) was designed to amplify and detect DNA of six bacterial respiratory pathogens. This assay was compared with real-time PCR assays based upon the same target sequences for the ability detect the target bacteria by use of both stock strains and specimens from respiratory disease patients. The ResPlex I assay is somewhat less sensitive than real-time PCR assays but offe...

A multiplex PCR for the simultaneous detection of Tenacibaculum maritimum and Edwardsiella tarda in aquaculture.

A specific and sensitive multiplex PCR (mPCR) method was developed as a useful tool for the simultaneous detection of two important flatfish pathogens in marine aquaculture, Tenacibaculum maritimum and Edwardsiella tarda. In fish tissues, the average detection limit for these mPCR-amplified organisms was 2 × 10 ⁵ ± 0.2 CFU/g and 4 × 10 ⁵ ± 0.3 CFU/g, respectively. These values are similar or ev...

Development and application of a multiplex PCR assay for rapid detection of 4 major bacterial pathogens in ducks.

Infections with Pasteurella multocida, Salmonella enterica, Riemerella anatipestifer, and Escherichia coli result in high morbidity and mortality, which cause significant economic loss in the poultry industry. It can be difficult to distinguish these pathogens based on clinical signs because these pathogens can cause similar clinical signs and coinfections can occur. Thus, rapid and sensitive d...
